Abstract

The study investigated an approach to enhancing the antibiofilm activity of surfactants produced by Rhodococcus erythropolis IMV Ac-5017 through their synthesis in the presence of the yeast inducer Saccharomyces cerevisiae BTM-1 in different physiological states (live and inactivated cells, as well as the corresponding supernatant) and their subsequent application in combination with tea tree essential oil. It was shown that, regardless of the inducer state, the obtained surfactants combined with the essential oil exhibited an enhanced ability to disrupt dual-species bacterial biofilms compared with the individual components and mixtures of oil with surfactants synthesized without an inducer. The degree of destruction of Escherichia coli IEM-1 and Staphylococcus aureus BMS-1 biofilms reached 75–81%, while that of Bacillus subtilis BT-2 and Pseudomonas sp. MI-2 biofilms reached up to 98%, significantly exceeding the effectiveness of the corresponding single agents. The obtained results confirm the feasibility of a biotechnological approach that combines controlled induction of microbial surfactant synthesis with their integration into formulations containing natural antimicrobial compounds as an effective strategy for controlling mixed biofilms formed by opportunistic microorganisms. У роботі досліджено підхід до підвищення антибіоплівкової активності поверхнево-активних речовин (ПАР) Rhodococcus erythropolis IMB Ac-5017 шляхом їх синтезу за наявності дріжджового індуктора Saccharomyces cerevisiae БTM-1 у різному фізіологічному стані (живі та інактивовані клітини, а також відповідний супернатант) та подальшого застосування у комплексі з ефірною олією чайного дерева. Показано, що незалежно від стану індуктора отримані ПАР у поєднанні з ефірною олією проявляють підвищену здатність до руйнування двовидових бактеріальних біоплівок порівняно з окремими компонентами та сумішами олії з поверхнево-активними речовинами, синтезованими без індуктора. Встановлено, що ступінь деструкції біоплівок Escherichia coli IEM-1 з Staphylococcus aureus BMS-1 досягав 75–81%, а для біоплівки Bacillus subtilis BT-2 з Pseudomonas sp. MI-2 — до 98% тощо, що істотно перевищувало показники дії монопрепаратів. Отримані результати підтверджують доцільність використання біотехнологічного підходу, що поєднує керовану індукцію синтезу мікробних ПАР та їх комбінування з природними антимікробними сполуками як ефективної стратегії контролю змішаних біоплівок умовно-патогенних мікроорганізмів.
