Abstract

Мета досліджень – обґрунтувати шляхи підвищення технічної ефективності машин роторного типу для пресування таблеток із врахуванням показників якості готової продукції та конструктивних і режимних параметрів роботи модуля пресування. Технічну ефективність пресу поряд з іншими факторами визначають якісні показники готової продукції – таблеток, а саме їх міцність, стираність, зовнішній вигляд, руйнування їх структури. Згадані показники залежать від густини таблетки, яка у свою чергу залежить від ступеню пресування та законів руху пуансонів, режимів роботи обладнання та стану поверхні пресувального інструменту. Шляхи регулювання технічної ефективності пресувальних машин у процесі роботи з урахуванням показників якості таблеток потребують обґрунтування та детальних досліджень. Об’єкт досліджень – модуль машини роторного типу для реалізації процесу пресування під час виготовлення таблеток. Предметом досліджень є функціональний зв’язок між конструктивними, режимними параметрами роботи модуля пресування, силовими та міцністними характеристиками деталей та складальних одиниць, якісними показниками таблеток і показниками технічної ефективності пресувальних машин роторного типу для виготовлення таблеток. Значущі фактори, які впливають на якість готової продукції та технічну ефективність пресу, визначені шляхом експертного опитування фахівців з таблетування та експлуатації обладнання за загальноприйнятими галузевими методами. Параметри технічної ефективності модуля пресування визначалася шляхом узагальнення відносних значень експертних оцінок і числових значень показників якості таблеток. Експериментальні дослідження проведені шляхом польового експерименту у виробничих умовах на прикладі роторної таблеткової машини Korsch XL, яка містить 63 комплекти пресувального інструменту, а частота обертання роторного стола становить від 20 до 80 об/хв. Аналіз показників якості проводився на прикладі зразків таблеток на основі мікрокристалічної целюлози і лактози. Відновлення поверхні пресувального інструменту проводилося за допомогою набору полірувального інструменту, і машинним методом шляхом оброблення пуансонів у абразивній полірувальній суміші. Стан поверхні пресувального інструменту визначався порівнянням із зразками шорсткості. Якісні показники таблеток визначалися фармакопейними методами: за міцність таблеток приймалося руйнівне напруження під час стискання таблетки у діаметральному напрямку, стираність таблеток визначалася як відсоток втрати їх маси у обертовому барабані. Розподіл густини в об'ємі таблетки аналізували методом симуляційного моделювання у програмному пакеті COMSOL Multiphysics®, який базується на методі скінченних елементів (МСЕ). Опитування експертів виконано з метою ранжування факторів таблетування за рівнем ризику для якості таблеток. Застосовано систематичний метод аналізу видів наслідків та відмов. Розглянуто 20 факторів, які впливають на процес таблетування за категоріями – властивості матеріалу, параметри процесу, механічні фактори, технологічні фактори, фактори впливу навколишнього середовища. Інтегральний показник ризику оцінено як добуток оцінок за серйозність наслідків, частоту виникнення та ймовірність виявлення проблеми. До категорії високого ризику віднесено 7 факторів, середнього – 11, низького – 2. Основними факторами визначено частоту обертання роторного стола та час витримування таблетки під тиском, густина таблетки, кількість циклів пресування. Розподіл дефектів має виражений Парето-характер: відколювання кришки, розтріскування, налипання гранул на пуансон і варіація ваги становлять 58,2%, якщо врахувати варіацію міцності та розшарування – 81,8%, тоді як подвійне пресування залишається граничним випадком 1,8%. На міцність та стираність таблетки впливають її густина, і, відповідно, закон руху пуансонів, час витримування таблетки між пуансонами, і, відповідно – частота обертання роторного стола і діаметр плоскої частини головки пуансона, стан поверхні пресувального інструменту, який залежить від часу роботи пресу. Така кореляція якісних характеристик таблетки із конструктивними та режимними параметрами роботи модуля пресування врахована у подальших дослідженнях. Параметри технічної ефективності пресувальної машини роторного типу розраховано з врахуванням факторів, які визначають якість готової продукції, а саме міцності, стираності, зовнішнього стану, наявності сколів і відхилення від заданої маси. Дані показники регулюються як рецептурою продукту та способом виготовлення грануляту, так і регулюванням роботи модуля пресування, зокрема: закону руху пуансонів та ступеню пресування для регулювання густини таблетки, швидкості обертання роторного стола, яка впливає на стираність і міцність таблетки, а також на відхилення від заданої маси; крім того, на показники якості впливає шорсткість робочої поверхні пресувального інструменту. Значне підвищення стираності таблетки після 250 тисяч циклів пресування знижує технічну ефективність пресу з 1,0 до 0,6. Незначно підняти технічну ефективність можливо за рахунок зниження частоти обертання роторного стола або ж відновити початковий показник ефективності шляхом полірування пресувального інструменту. Після досягнення 300 тисяч циклів пресування прес варто зупинити, провести очищення і полірування пресувального інструменту. Після полірування показники технічної ефективності машини підвищуються до початкового рівня у разі машинного полірування пуансонів, або не досягають початкового рівня при ручному обробленні поверхні. При густині до 0,7 г/см³ таблетка має низьку міцність, руйнується у руках і є непридатною для наступних операцій нанесення покриття, пакування і транспортування. Виділяються три зони, де зміна міцності описується різними законами: І – міцність таблетки зростає пропорційно до її густини; ІІ – зі збільшенням густини таблетки понад 1 г/см³ міцність зростає за параболічною залежністю; ІІІ – при значенні густини понад 1,5 г/см³ міцність таблетки знижується, і виникає такий вид браку як розшарування. Зі збільшенням густини стираність таблетки знижується за параболічною залежністю. При густині 0,8–1,0 г/см³ стираність досліджуваних таблеток стає меншою за 1%, що відповідає регламентованим значенням. Зі збільшенням густини понад 1,3–1,7 г/см³ стираність таблеток починає зростати за параболічною залежністю і стає більшою за регламентовані межі. Режимні параметри роботи таблетпресу впливають на міцність таблетки. У межах робочого діапазону частоти обертання барабану таблетпресу від 30 до 80 об/хв міцність таблеток може знизитися до 10%. Це пояснюється зміною швидкості пресування і зниженням часу витримування таблетки у матриці. Зі збільшенням частоти обертання барабану таблетпресу від 30 до 80 об/хв стираність таблетки збільшується від 0,2 до 1,2%, що більше за регламентовані фармакопейні (1%) та рекомендовані (0,5) норми. Це пояснюється зниженням часу витримування таблетки у матриці та більшою швидкістю виштовхування, що знижує міцність зовнішніх шарів таблетки. Кількість циклів пресування у прес-парі матриця–пуансон впливає на міцність таблетки: через 300 тисяч циклів таблетка втрачає в середньому до 8% міцності, а стираність зростає у декілька разів та перевищує регламентовані показники. Це пояснюється налипанням продукту на поверхні матриці і пуансона, збільшенням шорсткості їх поверхні і, відповідно, збільшенням коефіцієнта тертя і міцності адгезії. Внаслідок цього зовнішній шар таблетки має нижчі показники міцності та інтенсивно руйнується. Результати досліджень із визначення якісних показників таблеток узагальнені у вигляді рівняння інтерполяційної моделі другого порядку, яка описує вплив густини таблетки, частоти обертання роторного стола і кількості циклів пресування на міцність таблетки. Дане рівняння дозволяє передбачити міцність таблетки під час регулювання технічної ефективності преса шляхом регулювання частоти обертання роторного стола або відновлення стану поверхні пуансонів. Збільшення показників міцності та стираності накладає обмеження на частоту обертання ротора таблетпресу. Для використаних у дослідженнях таблеток і таблетпресу Korsch XL раціональна частота обертання ротора – 50 об/хв. Збільшення частоти обертання, і, відповідно, продуктивності, є можливим за рахунок частішого полірування поверхні матриць і пуансонів. При односторонньому пресуванні на протилежних краях таблетки у зоні контакту з поверхнею матриці виникають зони із більшими і меншими напруженням і густиною. Проте відносна густина у ядрі таблетки не перевищує середню густину більше ніж на 7%. Така неоднорідність густини незначно впливає на показники міцності і стираності, проте є ймовірною причиною розшарування і відколювання частинок таблетки. При двосторонньому пресуванні на краях таблетки у зоні контакту з матрицею виникають концентрації напружень і зони з більшою густиною. Густина у ядрі таблетки не перевищує середню густину більше ніж на 4%, що є меншим порівняно із таблеткою, виготовленою одностороннім способом пресування. Такий же результат підтверджено при конструктивно можливому двосторонньому способі пресування із синусоїдальним законом переміщення та прискорення пуансона. Внаслідок повільної швидкості на кінцевому етапі пресування відбувається релаксація напружень у таблетці, а густина частково вирівнюється по об’єму таблетки. Результати дослідження не підтверджують гіпотези про те, що спосіб пресування і закон руху пуансона значно впливають на розподіл густини і міцності у таблетці за умови, що її висота значно менша за діаметр. Силовий аналіз роботи вузла пресування проведено з метою визначення зусилля, крутного моменту і потужності на подолання опору руху пуансонів і, відповідно, роторного стола під час всіх циклів виготовлення таблетки – її попереднього і основного пресування та виштовхування. Для розрахунку силових характеристик вузла пресування використано дані експериментальних досліджень, які встановлюють вплив густини таблетки на зусилля основного і попереднього пресування. Зусилля пресування зростає зі зміною густини за параболічною залежністю. Звернено увагу, що двопотоковий спосіб пресування хоча і потребує у два рази більших енерговитрат, проте створює більш сприятливий для міцності і довговічності роботи деталей і вузлів режим, так як нерівномірність потужності менша порівняно із випадком однопотокового пресування – 59 і 88% відповідно. Встановлено, що під час контакту головки пуансона з пресувальним роликом внаслідок зміщення векторів зусилля пресування таблетки і зусилля притискання ролика на пуансон виникає момент сили, який призводить до збільшення напружень у зоні контакту пуансона з поверхнею напрямної втулки. Для пуансонів класу D, які мають більший діаметр плоскої частини головки, для уникнення значних напружень у зоні контакту з матрицею внаслідок перекосу посадковий діаметр має бути збільшений, і, відповідно, передбачений більший діаметр втулки. Тривалість полірування поверхні пуансонів машинним способом у абразивній полірувальній суміші до рівня шорсткості 1 мкм становить 54 хв порівняно із 165 хв при ручному поліруванні. Після машинного полірування технічна ефективність пресу відновлюється до початкових значень, а при ручному – відновлюється частково. Результати роботи рекомендовані до впровадження у виробничій діяльності АТ «Інженерія» та впроваджені у навчальний процес у вигляді лабораторних робіт, теоретичного матеріалу та опису конструкцій обладнання у навчальній літературі.
The aim of the research is to justify the ways to increase the technical efficiency of rotary-type machines for tablet compession, taking into account the quality indicators of the finished product and the design and operating parameters of the compessing module. The technical efficiency of the press, along with other factors, is determined by quality indicators of the finished products – tablets, namely their strength, friability, appearance, and structural failure. These indicators depend on tablet density, which in turn depends on the degree of compression and the laws of punch motion, the operating modes of the equipment, and the surface condition of the compression tooling. Ways to regulate the technical efficiency of compression machines during operation, taking into account tablet quality indicators, require substantiation and detailed investigation. The object of research is a rotary-type machine module for implementing the compressing process during the manufacture of tablets. The subject of research is the functional relationship between the design, operating parameters of the compressing module, the power and strength characteristics of parts and assembly units, the quality indicators of tablets and the technical efficiency indicators of rotary-type pressing machines for tablets manufacturing. Significant factors that affect finished product quality and the technical efficiency of the press were determined through an expert survey of specialists in tableting and equipment operation using generally accepted industry methods. The technical efficiency parameters of the compression module were determined by generalizing the relative values of expert assessments and the numerical values of tablet quality indicators. Experimental studies were carried out through a field experiment under production conditions using a Korsch XL rotary tablet press, which contains 63 sets of compression tooling, and the rotational speed of the rotary table ranges from 20 to 80 rpm. Quality indicators were analyzed using tablet samples based on microcrystalline cellulose and lactose. The surface of the compression tooling was restored using a polishing tool kit and by a machine method through processing punches in an abrasive polishing mixture. The surface condition of the compression tooling was determined by comparison with roughness specimens. Tablet quality indicators were determined by pharmacopoeial methods: tablet strength was taken as the breaking stress during diametral compression of the tablet, while tablet friability was determined as the percentage loss of mass in a rotating drum. The density distribution within the tablet volume was analyzed by simulation modelling in the COMSOL Multiphysics® software package, which is based on the finite element method (FEM). The expert survey was conducted to rank tableting factors by their level of risk to tablet quality. A systematic failure mode and effects analysis method was applied. Twenty factors affecting the tableting process were considered by category: material properties, process parameters, mechanical factors, technological factors, and environmental impact factors. The integral risk indicator was evaluated as the product of scores for severity of effects, occurrence frequency, and probability of detecting the problem. Seven factors were assigned to the high-risk category, 11 to the medium-risk category, and 2 to the low-risk category. The main factors were identified as the rotational speed of the rotary table and the tablet dwell time under pressure, tablet density, and the number of compression cycles. The distribution of defects has a pronounced Pareto pattern: capping, cracking, granule sticking to the punch, and weight variation account for 58.2%; when strength variation and lamination are included, the share reaches 81.8%, while double compression remains a limiting case at 1.8%. Tablet strength and friability are affected by tablet density and, accordingly, the law of punch motion; the dwell time of the tablet between the punches and, accordingly, the rotational speed of the rotary table and the diameter of the flat part of the punch head; and the surface condition of the compression tooling, which depends on the press operating time. This correlation between tablet quality characteristics and the design and operating parameters of the compression module was taken into account in further studies. The technical efficiency parameters of a rotary-type compression machine were calculated taking into account the factors that determine finished product quality, namely strength, friability, external condition, the presence of chips, and deviation from the specified mass. These indicators are regulated both by the product formulation and the granulate manufacturing method, and by adjustment of the compression module operation, in particular: the law of punch motion and degree of compression for regulating tablet density; the rotational speed of the rotary table, which affects tablet friability and strength, as well as deviation from the specified mass; in addition, the roughness of the working surface of the compression tooling affects the quality indicators. A significant increase in tablet friability after 250 thousand compression cycles reduces the technical efficiency of the press from 1.0 to 0.6. The technical efficiency can be slightly increased by reducing the rotational speed of the rotary table, or the initial efficiency indicator can be restored by polishing the compression tooling. After reaching 300 thousand compression cycles, the press should be stopped, and the compression tooling should be cleaned and polished. After polishing, the technical efficiency indicators of the machine increase to the initial level in the case of machine polishing of the punches, or do not reach the initial level in the case of manual surface treatment. At a density of up to 0.7 g/cm³, the tablet has low strength, disintegrates in the hands, and is unsuitable for subsequent coating, packaging, and transportation operations. Three zones are distinguished in which the change in strength is described by different laws: Zone I: Tablet strength increases in proportion to its density. Zone II: With an increase in tablet density above 1 g/cm³, strength increases according to a parabolic relationship. Zone III: Decrease in strength. At a density value above 1.5 g/cm³, tablet strength decreases, and defects such as lamination occur. As density increases, tablet friability decreases according to a parabolic relationship. At a density of 0.8–1.0 g/cm³, the friability of the studied tablets becomes less than 1%, which corresponds to the regulated values. With an increase in density above 1.3–1.7 g/cm³, tablet friability begins to increase according to a parabolic relationship and becomes higher than the regulated limits. The operating parameters of the tablet press affect tablet strength. Within the operating range of the rotary table rotational speed from 30 to 80 rpm, tablet strength may decrease by up to 10%. This is explained by a change in compression speed and a decrease in the dwell time of the tablet in the die. With an increase in the rotary table rotational speed from 30 to 80 rpm, tablet friability increases from 0.2 to 1.2%, which is higher than the regulated pharmacopoeial limit (1%) and the recommended limit (0.5%). This is explained by a decrease in tablet dwell time in the die and a higher ejection speed, which reduces the strength of the outer tablet layers. The number of compression cycles in the die-punch compression pair affects tablet strength: after 300 thousand cycles, the tablet loses on average up to 8% of its strength, while friability increases severalfold and exceeds the regulated indicators. This is explained by product sticking to the surfaces of the die and punch, an increase in their surface roughness, and, accordingly, an increase in the coefficient of friction and adhesion strength. As a result, the outer layer of the tablet has lower strength indicators and undergoes intensive failure. The results of studies on determining tablet quality indicators were generalized in the form of an equation of a second-order interpolation model, which describes the effect of tablet density, rotary table rotational speed, and the number of compression cycles on tablet strength. This equation makes it possible to predict tablet strength when regulating the technical efficiency of the press by adjusting the rotational speed of the rotary table or restoring the surface condition of the punches. An increase in strength and friability indicators imposes restrictions on the rotational speed of the tablet press rotary table. For the tablets and Korsch XL tablet press used in the studies, the rational rotary table speed is 50 rpm. Increasing the rotational speed and, accordingly, productivity is possible through more frequent polishing of the surfaces of the dies and punches. During single-sided compression, zones with higher and lower stress and density occur on opposite tablet edges in the contact zone with the die surface. However, the relative density in the tablet core does not exceed the average density by more than 7%. Such density nonuniformity has a minor effect on strength and friability indicators, but it is a probable cause of lamination and chipping of tablet particles. During double-sided compression, stress concentrations and zones with higher density occur at the tablet edges in the contact zone with the die. The density in the tablet core does not exceed the average density by more than 4%, which is lower compared with a tablet manufactured by the single-sided compression method. The same result was confirmed for the structurally feasible double-sided compression method with a sinusoidal law of punch displacement and acceleration. Due to the low velocity at the final stage of compression, stress relaxation occurs in the tablet, and the density is partially equalized throughout the tablet volume. The research results do not confirm the hypothesis that the compression method and the law of punch motion significantly affect the distribution of density and strength in the tablet, provided that its height is much smaller than its diameter. The force analysis of the compression unit operation was carried out to determine the force, torque, and power required to overcome the resistance to punch motion and, accordingly, the resistance of the rotary table during all cycles of tablet manufacturing – pre-compression, main compression, and ejection. To calculate the force characteristics of the compression unit, experimental study data were used that establish the effect of tablet density on the main and pre-compression forces. The compression force increases with changes in density according to a parabolic relationship. It was noted that the two-stream compression method, although it requires twice as much energy consumption, creates more favorable conditions for the strength and service life of parts and units, since power nonuniformity is lower compared with the case of single-stream compression – 59% and 88%, respectively. It was established that during contact between the punch head and the compression roller, due to the displacement of the vectors of the tablet compression force and the roller pressing force on the punch, a force moment arises, which leads to an increase in stresses in the contact zone between the punch and the surface of the guide bushing. For class D punches, which have a larger diameter of the flat part of the head, the seating diameter must be increased to avoid significant stresses in the contact zone with the die due to skewing, and, accordingly, a larger bushing diameter must be provided. The duration of machine polishing of punch surfaces in an abrasive polishing mixture to a roughness level of 1 μm is 54 min compared with 165 min for manual polishing. After machine polishing, the technical efficiency of the press is restored to the initial values, whereas after manual polishing it is restored partially. The results of the work are recommended for implementation in the production activities of JSC “Engineering” and have been implemented in the educational process in the form of laboratory lessons, theoretical material, and descriptions of equipment designs in educational literature.
